Subsequently, the DRD2 gene had been studied in many species, including human, rat, turkey, canine, pig and chicken (Gandelman et al., 1991; Grandy et al., 1989; Itokawa et al., 1993; Montmayeur et al., 1991; Myeong et al., 2000; Schnell et al., 1999; Seeman et al., 2000; Toso et al., 1989; Xu et al., 2010, 2011). In turkeys, two spliced isoforms were isolated and some studies suggested that the expression of the DRD2 mRNA was correlated with broodiness (Schnell et al., 1999).

Though Ser311/Cys311 has been proposed as a genetic risk factor for schizophrenia following the observation of an excess of Ser311/Cys311 variants among schizophrenics, a number of replications failed to confirm the initial finding (Arinami et al., 1994; Chen et al., 1996; Laurent et al., 1994).

This S311C mutation (NCBI SNP ID rs1801028) (37) is present at third cytoplasmic loop of the human D2 DA receptor, which is responsible for effective signal transduction of these receptors (41, 42). It is further reported that D2 DA receptor S311C allelic variants do not severely affect ligand binding of the receptor and its coupling with the G protein; rather, the receptor variant becomes significantly less effective than the wild type in relation to lower efficiency in activating the α subunit of the G protein heterotrimer to transduce downstream signals (41, 42).
