Identifying the human aggrecanase
2010, Osteoarthritis and CartilageCitation Excerpt :Given the differences between the aetiopathogenesis of hip and knee OA19, it is not surprising that ADAMTS expression differs between knee and hip joints in humans15 and presumably differs also with stage of disease, although this has not been systematically examined. ADAMTS expression in cartilage represents only one part of the total joint picture, since ADAMTS enzymes are expressed in joint capsule and meniscus, as well as in synovium where ADAMTS expression is particularly well documented20–28. In distinct contrast to cartilage, both ADAMTS-4 and -5 are expressed constitutively in synovium and their expression by synovial fibroblasts is not regulated by IL-1 in vitro20,21,29.
Co-culture of mechanically injured cartilage with joint capsule tissue alters chondrocyte expression patterns and increases ADAMTS5 production
2009, Archives of Biochemistry and BiophysicsCitation Excerpt :Nevertheless, it is possible that tissue from mature animals will behave differently in these injury models and this should be considered when making comparisons to other studies. Both synovium and joint capsule tissue generate a soluble aggrecan-degrading activity in vitro[30,31]. Higher aggrecanase-mediated cleavage potency was measured in medium from co-culture of synovium with either live or dead cartilage than that seen from synovium alone [30].
Western blot quantification of aggrecan fragments in human synovial fluid indicates differences in fragment patterns between joint diseases
2009, Osteoarthritis and CartilageCitation Excerpt :Secondly, joint cartilage is a major source of aggrecan fragments appearing in SF, but may not be the sole source; they may also to a variable extent originate from, e.g., menisci, ligaments and synovial tissue39–41. The synovial cells of the joint capsule contain aggrecanase activities42,43. Thirdly, we have assumed that aggrecan fragments appearing in SF are the result of proteolysis of resident, functional matrix molecules.
Effects of glucosamine on proteoglycan loss by tendon, ligament and joint capsule explant cultures
2008, Osteoarthritis and CartilageCitation Excerpt :In addition there was no effect on the proteolytic processing of large proteoglycans (data not shown). ADAMTS-4 and-5 are expressed in joint fibrous connective tissues and one or both of these proteinases may be responsible for the degradation of large aggregating proteoglycans in these tissues2,4,18,20,40–43. The lack of effect of glucosamine treatment in joint fibrous connective tissue explants is likely to be due to the difference in the regulation of aggrecanase activity in these tissues compared to cartilage.
