  1. 1.

    The uptake of threonine, isoleucine, valine and leucine, their degradation to higher alcohols, and the regulation of the reactions involved were investigated using an auxotrophic mutant ofSaccharomyces cerevisiae, mating type a, genetic markers ade2, hom2, thr4, ilv2, leu1. The formation of higher alcohols from amino acids by the Ehrlich pathway is restricted to the exponential growth phase. No formation of higher alcohols occurs in the stationary phase although a further consumption of amino acids is observed. The synthesis of higher alcohols by the auxotrophic yeast strain is directly proportional to the corresponding amino acids in the medium. Increasing concentrations of glucose slightly inhibit the uptake of amino acids and their degradation to higher alcohols.

    The yeast cells excrete amino acids during growth and in the stationary phase, presumably alanine, glutamic acid, arginine, lysine, proline, and serine. Aspartic acid, cystine, tyrosine, phenylalanine, tryptophane, 4-aminobutyric acid, and ornithine appear in the fermented medium in small amounts. The excretion of amino acids is increased by a high nitrogen content of the medium.

    Die Aufnahme und der Abbau von Threonin, Isoleucin, Valin und Leucin zu höheren Alkoholen, die Regulation der hieran beteiligten Reaktionen wurde bei einer Mutante vonSaccharomyces cerevisiae untersucht. Der Abbau von Aminosäuren zu höheren Alkoholen ist auf die exponentielle Wachstumsphase begrenzt. Während der stationären Phase werden keine höheren Alkohole mehr gebildet, obwohl sich eine weitere Aufnahme der Aminosäuren beobachten läßt. Die Produktion höherer Alkohole ist dem Angebot der entsprechenden Aminosäuren proportional. Die Aufnahme der Aminosäuren und deren Abbau zu höheren Alkoholen wird durch steigende Glucosemengen geringfügig gehemmt.

    Während des aktiven Wachstums und der stationären Phase werden Aminosäuren ausgeschieden, vor allem Alanin, Glutaminsäure, Arginin, Lysin, Prolin und Serin. In geringen Mengen werden Asparaginsäure, Cystin, Tyrosin, Phenylalanin, Tryptophan, 4-Aminobuttersäure und Ornithin gefunden. Die Ausscheidung ist dem Stickstoffangebot proportional.
