Rapid colorimetric assay for cell growth and survival: Modifications to the tetrazolium dye procedure giving improved sensitivity and reliability
Abstract
A convenient way to estimate the number of viable cells growing in microtitre tray wells is to use a colorimetric assay and an automatic microplate scanning spectrophotometer. One such assay, developed by Mosmann, depends on the reduction by living cells of tetrazolium salt, MTT, to form a blue formazan product. However the original technique has several technical limitations, namely a less than optimal sensitivity, a variable background due to protein precipitation on adding an organic solvent to dissolve the blue formazan product, and a low solubility of the product. These problems have been overcome by the following modifications: avoidance of serum in the incubation medium, thus overcoming precipitation problems in the organic solvent; avoidance of phenol red in the incubation medium, thus avoiding the use of acid in the final solvent which altered the spectral properties of the formazan; elimination of the medium containing MTT after the reaction and subsequent use of pure propanol or ethanol to rapidly solubilize the formazan; use of a higher concentration of MTT; use of half-area microtitre trays to increase the spectrophotometer readings from a given amount of formazan; use of a more judicious reference wavelength in a dual wavelength spectrophotometer. With these modifications the reliability and sensitivity of the test have been increased to the point where it can in many cases replace the [3H]thymidine uptake assay to measure cell proliferation or survival in growth factor or cytotoxicity assays. Examples of its use in IL-2 assays are given.

Studies on the cytotoxicity of leaf protein concentrate from Diplazium esculentum, pasta preparation and assessment of its quality during storage
2024, Food Chemistry AdvancesCytotoxicity studies performed with the cell lines viz., HEK 293 (Human embryonic kidney cell line), HEP G2 (Hepatoblastoma cell line) and PBMC (Peripheral blood mononuclear cells) by leaf protein concentrate (LPC) of Diplazium esculentum were analysed with an aim to possibly determine the safety of leaf protein concentrate. Results revealed the safety from the cytotoxic point of view at various levels of incorporation into pasta such as 5 %, 10 %, 15 % and 20 %. Scanning electron microscopy revealed that gluten network in which starch granules were embedded, control pasta had a smooth surface whereas leaf protein concentrate incorporated pasta had a more open structure is apparent showing the protein matrix with the majority of the starch granules absent and it was further supported by textural analysis. The proximate composition, cooking quality parameters chronologically increased with increase in percentage of leaf protein concentrate in the pasta. Aerobic plate count, yeast and mold count of pasta were within the permissible limit. Significant variations in microbial load were seen throughout the course of the same storage period when LPC was included at various quantities. After the first 30 days of storage, yeast and mold started to grow, but they did so within the allowed limits. Pasta with a 20 % addition of protein concentrate was optimised for consumption and may be safely consumed for up to 30 days. According to this study, leaf protein concentrate may be delivered in pasta-like goods to increase their nutraceutical potential without compromising the products' sensory quality or safety.
Mechanism of surfactant interactions with feline coronavirus: A physical chemistry perspective

Yet, the detailed mechanisms by which surfactants deactivate coronaviruses remain underexplored. This study delves into the virucidal mechanisms of various surfactants on Feline Coronavirus (FCoV) and their potential applications against more pathogenic coronaviruses.
By integrating virucidal activity assays with fluorescence spectroscopy, dynamic light scattering and laser Doppler electrophoresis, alongside liposome permeability experiments, we have analyzed the effects of non-ionic and ionic surfactants on viral activity.
The non-ionic surfactant octaethylene glycol monodecyl ether (C10EO8) inactivates the virus by disrupting the lipid envelope, whereas ionic surfactants like Sodium Dodecyl Sulfate and Cetylpyridinium Chloride predominantly affect the spike proteins, with their impact on the viral membrane being hampered by kinetic and thermodynamic constraints. FCoV served as a safe model for studying virucidal activity, offering a faster alternative to traditional virucidal assays. The study demonstrates that physicochemical techniques can expedite the screening of virucidal compounds, contributing to the design of effective disinfectant formulations. Our results not only highlight the critical role of surfactant-virus interactions but also contribute to strategic advancements in public health measures for future pandemic containment and the ongoing challenge of antimicrobial resistance.
Ginseng root extract-mediated synthesis of monodisperse silver nanoparticles as a fluorescent probe for the spectrofluorometric determination of nilvadipine; Evaluation of remarkable anti-bacterial, anti-fungal and in-vitro cytotoxic activities
2024, Spectrochimica Acta - Part A: Molecular and Biomolecular SpectroscopyNanoparticles are a boon for humanity because of their improved functionality and unlimited potential applications. Considering this significance, the proposed study introduced a simple, fast and eco-friendly method for synthesis of fluorescent silver nanoparticles (Ag-NPs) using Panax Ginseng root extract as a reducing and capping agent. Synthesis of Ag-NPs was performed in one step within three minutes utilizing microwave irradiation. The resulting Ag-NPs were characterized using various microscopic and spectroscopic techniques such as, Transmission Electron Microscope (TEM), UV/Visible spectroscopy, Fourier Transform Infrared Spectroscopy (FTIR) and Energy Dispersive X-ray analysis (EDX). The prepared Ag-NPs, which act as a fluorescent nano-probe with an emission band at 416 nm after excitation at 331 nm, were used to assay nilvadipine (NLV) spectrofluorimetrically in its pharmaceutical dosage form with good sensitivity and reproducibility. The proposed study is based on the ability of NLV to quantitatively quench the native Ag-NPs fluorescence, forming a ground state complex as a result of static quenching and an inner filter mechanism. The suggested approach displayed a satisfactory linear relationship throughout a concentration range of 5.0 μM − 100.0 μM, with LOD and LOQ values of 1.18 μM and 3.57 μM, respectively. Validation of the suggested approach was examined in accordance with ICH recommendations. In addition, the anti-bacterial and anti-fungal activities of the prepared nanoparticles were investigated, and they demonstrated effective anti-microbial activities and opened a future prospective to combat future antibiotic resistance. Finally, in-vitro cytotoxicity assay of Ag-NPs against normal and cancerous human cell lines was studied using MTT assay. The results proved the potential use of the produced Ag-NPs as an adjunct to anticancer treatment or for drug delivery without significantly harming healthy human cells.
Novel 6-hydroxybenzothiazol-2-carboxamides as potent and selective monoamine oxidase B inhibitors endowed with neuroprotective activity
2024, European Journal of Medicinal ChemistryIn neurodegenerative diseases, using a single molecule that can exert multiple effects to modify the disease may have superior activity over the classical “one molecule-one target” approach. Herein, we describe the discovery of 6-hydroxybenzothiazol-2-carboxamides as highly potent and selective MAO-B inhibitors. Variation of the amide substituent led to several potent compounds having diverse side chains with cyclohexylamide 40 displaying the highest potency towards MAO-B (IC50 = 11 nM). To discover new compounds with extended efficacy against neurotoxic mechanisms in neurodegenerative diseases, MAO-B inhibitors were screened against PHF6, R3 tau, cellular tau and α-synuclein (α-syn) aggregation. We identified the phenethylamide 30 as a multipotent inhibitor of MAO-B (IC50 = 41 nM) and α-syn and tau aggregation. It showed no cytotoxic effects on SH-SY5Y neuroblastoma cells, while also providing neuroprotection against toxicities induced by α-syn and tau. The evaluation of key physicochemical and in vitro-ADME properties revealed a great potential as drug-like small molecules with multitarget neuroprotective activity.
A novel paddlewheel-like complex of [Pd(SCN)2(3-Acpy)2], (1) was produced at ambient temperature by adding an aqueous solution of palladium chloride to an aqueous acetonitrile solution of 3-acetylpyridine (3-Acpy) and potassium thiocyanate. Elemental analysis, Fourier-transform infrared (FT-IR), ultraviolet–visible (UV–vis), and nuclear magnetic resonance (NMR) spectroscopic methods were used to characterize complex 1 comprehensively. Single crystal X-ray diffraction was used to analyze the crystal structure of complex 1. The palladium atom is bonded to two 3-Acpy ligands and two thiocyanates groups in trans mode giving square planer geometry in complex 1. The discrete units of paddlewheel-like complex 1 through hydrogen bonding extend to form a 3D supramolecular network structure. The synthesized and characterized new palladium complex 1 was utilized as a precatalyst for the cyanation of alkyl bromide using potassium ferrocyanide as a benign cyanide source reagent. The suggested catalytic test reaction under high-pressure conditions via a Q-tube system in green solvent cyclopentyl methyl ether introduced high yields and a short reaction time. Moreover, the cytotoxic activity of complex 1 was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay against HePG-2 of liver cancer and MCF7 of breast cancer cell lines. According to the findings, complex 1 has significant strong activity against the cancer cell lines HePG-2 and MCF-7. Additionally, the luminescence spectra of complex 1 were investigated.
Design, synthesis, and anticancer evaluation of N-sulfonylpiperidines as potential VEGFR-2 inhibitors, apoptotic inducers
2024, Bioorganic ChemistryA new panel of N-sulfonylpiperidine derivatives has been designed and synthesized as vascular endothelial growth factor receptor-2 (VEGFR-2) inhibitors. Anti-proliferative activities of the synthesized members were tested against colorectal carcinoma (HCT-116), hepatocellular carcinoma (HepG-2), and breast cancer (MCF-7) cell lines. Compounds 3a, 4, 8, and 9 showed the highest activities against the tested cell lines. In particular, compound 8 showed excellent activities against HCT-116, HepG-2, and MCF-7 with IC50 values of 3.94, 3.76, and 4.43 μM, respectively. Such IC50 values are comparable to vinblastine (IC50 = 3.21, 7.35, 5.83 μM, respectively) and doxorubicin (IC50 = 6.74, 7.52, 8.19 μM, respectively). In vitro VEGFR-2 inhibitory activity of the most promising molecules (3a, 4, 8, and 9) indicated that compound 8 is the highest VEGFR-2 inhibitor with an IC50 of 0.0554 μM, compared to sorafenib (IC50 = 0.0416 μM). The most promising candidates (3a, 4, 8, and 9) were subjected to flow cytometry analyses to assess their effects on the cell cycle behavior and the apoptotic power against the three tested cell lines (HCT-116, HepG-2, and MCF-7). The tested compound arrested the tumor cells at both the G2/M and Pre-G1 phases. In addition, compound 9 was proved as the most effective apoptotic inducer among the tested compounds against the tested cells. Molecular docking studies against VEGFR-2 (PDB ID: 2OH4) revealed good binding modes of the synthesized compound similar to that of sorafenib. Computational investigation of ADMET parameters revealed the drug-likeness of the synthesized compounds.
