Characterization of the reverse transcriptase from a new retrovirus (HTLV) produced by a human cutaneous T-cell lymphoma cell line

Abstract

The reverse transcriptase from a new retrovirus, HTLV, isolated from a human cutaneous T-cell lymphoma cell line was characterized. The enzyme is associated with high-molecular-weight RNA of the virus particle, has a molecular weight of about 95,000, utilizes the same template primers (including natyral mRNAs) as other viral reverse transcriptases, and prefers magnesium as a divalent cation using various synthetic homopolymeric template primers. HTLV reverse transcriptase can be immunologically distinguished from those of known type C, B, and D retroviruses by enzyme activity neutralization tests. These and other results support the interpretation that HTLV is a novel oncornavirus which may be associated with certain human T-cell neoplasias.

      The supernatant was harvested after two days and frozen in aliquots. Reverse transcriptase (RT) activity in the supernatant was measured using 3H-labeled nucleotide triphosphates for initial viral production assays and 32P-labeled nucleotide triphosphates for all viral adaptation assays, by a previously described method (Rho et al., 1981). To produce the virus in the Cf2Th cell line, 100,000 Cf2-T4R5 cells were infected with 3000 cpm (3H) of HIV-1NL4.3(JR-FL) virus made in 293T cells as described above.

      Cell supernatants were collected at 72 h post-transfection, and were purified through a 0.45 µM syringe tip filter. The amount of virus in the supernatants was measured by reverse transcriptase (RT) assay [27]. Viruses harvested from the supernatant of the culture media were stored at −80 °C.
