Reverse transcriptase (RT) activity in the supernatant was measured using 3H-labeled nucleotide triphosphates for initial viral production assays and 32P-labeled nucleotide triphosphates for all viral adaptation assays, by a previously described method (Rho et al., 1981). To produce the virus in the Cf2Th cell line, 100,000 Cf2-T4R5 cells were infected with 3000 cpm (3H) of HIV-1NL4.3(JR-FL) virus made in 293T cells as described above.
