The antagonism of prostaglandin FP receptors inhibits the evolution of spreading depolarization in an experimental model of global forebrain ischemia
2020, Neurobiology of DiseaseCitation Excerpt :The administration of AL-8810 did not exert any discernable impact on the ramification of microglia processes (ramification index: 148.6 ± 63.8 and 138.3 ± 80.3, control and AL-8810) (Fig. 5D-E). FP receptors have been previously identified in brain synaptosomes and microvessels in newborn and adult pigs (Li et al., 1993, 1994), and cultured rat astrocytes (Kitanaka et al., 1994). Here we set out to visualize FP receptor presence on neuronal, glial (astrocyte and microglia) and cerebrovascular (endothelial and smooth muscle cell) compartments as potential sites of AL-8810 action.
Comprehensive profiling of prostaglandins in human ovarian follicular fluid using mass spectrometry
2018, Prostaglandins and Other Lipid MediatorsCitation Excerpt :The average estimated F2-isoprostane peak (RT 11.5 min) concentration is about 52 pg/ml or 150 pM. In general, PGF2α binding to its receptors occurs between 100 pM and 10 nM with reported dissociation constants in the high pM to low nM range, depending on the study [44–48]. An important mechanistic consideration comes from C. elegans ovaries, where numerous F-series PGs act collectively [27].
Prostaglandin FP receptor inhibitor reduces ischemic brain damage and neurotoxicity
2012, Neurobiology of DiseaseCitation Excerpt :Reports have suggested that prostaglandins perform diverse functions in neurodegenerative disorders through their G-protein‐coupled receptors (Doré, 2006). The FP receptor mRNA is expressed in rat whole brain (Kitanaka et al., 1994), and it has been reported that the FP receptor is expressed in cortical cultures enriched in neurons (Carlson, 2003). Activation of the FP receptor initiates several events, including stimulation of the inositol 1,4,5-trisphosphate receptor-related Ca2+ signaling pathway and store-operated Ca2+ entry (Snetkov et al., 2006).
