Mechanism of Bidirectional Leading-Strand Synthesis Establishment at Eukaryotic DNA Replication Origins
2019, Molecular CellCitation Excerpt :Elucidating the mechanism of leading-strand synthesis establishment represents an important advancement in our understanding of bidirectional DNA replication initiation at eukaryotic origins. Interestingly, despite significant compositional and structural differences existing between the prokaryotic, viral, and eukaryotic DNA replication machinery (O’Donnell et al., 2013), the use of lagging-strand primers to initiate bidirectional leading-strand replication now appears to be a general mechanism (Bullock et al., 1991; Fang et al., 1999; Hay and DePamphilis, 1982). We speculate that conservation of this process may simply result from the efficient priming required for discontinuous lagging-strand synthesis, negating the need for a dedicated leading-strand priming mechanism to initiate bidirectional DNA replication at origins.

2016, DNA RepairCitation Excerpt :It is likely that slipped-heteroduplex formation was increased when CTG repeats were in the lagging strand template because CTG repeats form biophysically more stable structures than CAG repeats. Slipped-DNA structures are likely more prone to form in the lagging strand template as it is left single-stranded for stretches of ∼300 nucleotides prior to the synthesis of Okazaki fragments [58,94]. We show that a deficiency of human ligase I, either as result of reduced catalytic activity or a defect in binding to PCNA, also increased the formation of slipped-DNAs, indicating that slipped-DNA formation occurs on the lagging strand, as LigI is critically required for efficient joining of Okazaki fragments [77–81].
