In general, TATA elements (located ∼25–30 bases upstream of the TSS) and an initiator motif (a pyrimidine-rich sequence encompassing the TSS) are important for eukaryotic gene expression (51–53). Inconsistently, however, our data showed that deletion of the region between bases −631 and −1 upstream of the mouse Insc TSS only partially decreased mouse Insc expression.

It has been observed that under mild stress conditions, the binding activity of HSF is also lessened to some extent, and it needs the synthesis of proteins especially the HSF itself (Zimarino et al., 1990; Wiederrecht et al., 1987; Parker and Topol, 1984). Stress conditions are necessary for the activation of HSF and its expression is constitutive in most tissues and cells, but it remains inactive in the absence of stress stimuli (Fiorenza et al., 1995).

In general, after cells receiving heat shock or other stresses, the Hsp70 gene has been shown to be activated by a transcription factor such as a heat shock factor (HSF), via binding of homotrimeric HSFs to upstream cis-elements [heat shock elements (HSEs)] in Hsp70 (Wu, 1984a,b, 1995; Parker and Topol, 1984; Topol et al., 1985; Abane and Mezger, 2010; Akerfelt et al., 2010; Fujimoto and Nakai, 2010; Sakurai and Enoki, 2010). In the present study, to understand further the gene activation mechanism of Hsp70a and Samui, we focused on the relationship between HSF and activation of these genes.
