Plakoglobin: A protein common to different kinds of intercellular adhering junctions

Abstract

We have established, by means of a monoclonal antibody and a cDNA clone, that a desmosomal polypeptide of Mr 83,000 also occurs at the plaques of other types of adhering junctions, including the vinculin-actin-associated intercellular junctions, e.g., the zonula adhaerens of epithelial cells and the endothelial, lens, and Sertoli cell junctions. This is the first component found in common among otherwise biochemically distinct plaque domains. Despite its concentration at these intercellular junctions, it is absent from the respective cell-substratum contact sites. In addition, it appears in a globular soluble 7S form in the cytoplasm. We discuss the significance of this protein, for which the name plakoglobin is proposed, in terms of its interaction with such biochemically diverse membrane domains and their different types of associated cytoskeletal filaments.

      The DESeq2 analysis identified 26 DEGs between RPCs stimulated with electro-chemotactic fields versus stimulation with SDF-1 fields, only. As seen in Table 3, 4 genes were upregulated in the RPCs exposed to electro-chemotactic fields: (1) Talin 2: A protein associated with focal adhesion formation that links integrins to the actin cytoskeleton (Critchley, 2000; Debrand et al., 2009); (2) lmo7: important in protein-protein interactions involved in ubiquination, shuttling of transcription factors (Holaska et al. (2006)) and retinal development (Semenova et al. (2003))(Ooshio et al., 2004); (3) JUP: a member of the catenin family that forms distinct complexes with cadherins and desmosomal cadherins linking them to the actin cytoskeleton (Cowin et al., 1986; Lampugnani et al., 1995); and (4) Svil: a protein tightly-associated with both actin filaments and plasma membranes, suggested for roles as a high-affinity link between the actin cytoskeleton and the membrane (Oh et al., 2003). This list of DEGs was then used for PAGE with the KEGG gene set database to determine enriched or overrepresented metabolic pathways.

      Our cellular model reflects that the control of spermatogenesis is induced by FSH in SC in vivo. PLKA is a constitutive protein of desmosomes and adherent junctions at the blood-testis barrier with structural and signalling roles (Cowin et al., 1986). PLKA interacts with junctional proteins such as desmosomal cadherins, classical cadherins and α-catenin.

      Notably, depletion of Ctnnb1 has no effect on hESC adhesion, although it is an integral component of adherens junctions (Kemler, 1993). Jup also participates in adherens junctions and has been shown to compensate for the loss of Ctnnb1 (Cowin et al., 1986; Lyashenko et al., 2011; Zhou et al., 2007). For example, in Ctnnb1-deficient mESCs, JUP levels are elevated and could rescue cell adhesion; therefore, knockdown of Jup in Ctnnb1 knockout mESCs disrupts adhesion (Lyashenko et al., 2011).
