Abstract

A method for quantitative estimation of glycerol and total carbohydrate in biological samples is described. The samples, deproteinized with cold acetone or trichloroacetic acid, were treated in the cold with 10 vols. of 0.75% (w/w) anthrone in 84% (w/w) sulphuric acid, and then heated 10 min at 100°C. Absorbance at 590 nm was used for evaluation of total carbohydrate content in the sample. The absorbance at 510 nm was used for the combined carbohydrate and glycerol estimation. This latter observation leads to the determination of the glycerol content of the sample because the carbohydrate interference is known from the data obtained at 590 nm. Uses of this method to determine glycerol and carbohydrate content in lipids of chicken egg-yolk samples are presented.

It was found that the plant grown in biochar had a total protein concentration of 3.39 mg/g of fresh leaf weight whereas pesticide enhanced the production of carbohydrate which was found to be 4.37 mg/g of fresh leaf weight as shown in Fig. 10a. The total carbohydrate in the plant leaf extract was estimated using Anthrone reagent (Pons et al., 1981) and maximum concentration was found in the control setup. It was found that the plant grown in biochar had carbohydrate concentration 0.578 mg/g of fresh leaf weight whereas; pesticide enhanced the production of carbohydrate which was found to be 0.78 mg/g of fresh leaf weight as shown in Fig. 10b.

  • Revealing the heating value characteristics of sludge-based hydrochar in hydrothermal process: from perspective of hydrolysate

    2021, Water Research
    Citation Excerpt :

A three-dimensional excitation-emission matrix (EEM) (F-4600, Hitachi) test was used to identify compounds in hydrolysate as described in SI. Protein and carbohydrate contents were quantified using Lowry and anthrone-based methods (Lowry et al., 1951; Pons et al., 1981). Chemical oxygen demand (COD) was used to quantify the concentration of organics in the hydrolysate.
