HLA class I expression of cell lines was measured by flow cytometry following surface staining using hybridoma supernatants for both anti-HLA-A2 (BB7.2 [HB-82 ; ATCC] (Parham and Brodsky, 1981); produced in-house) and pan-HLA class I (W6/32 [HB-95; ATCC] (Brodsky and Parham, 1982); produced in-house) as primary antibodies and goat anti-mouse IgG PE (Southern Biotech, Birmingham, AL, USA) as the secondary antibody. The cell lines were expanded to 1 × 109 in roller bottles, harvested by centrifugation (3724g, 15 min, 4 °C), snap frozen in liquid nitrogen and stored at −80 °C until required.
