2015, European Journal of PharmacologyCitation Excerpt :This will not only affect the luminal signal to promote protein folding, but most other cell signaling processes such as potassium channels (BK) supported Ca2+ sparks, spontaneous calcium-activated chloride currents (STICS), NAADP-initiated lysosomal Ca2+ signaling, mitochondrial oxidation and cytochrome c release, and a host of other cellular processes. Due to the all-important role of the ER/SR in cellular Ca2+ signaling (Berridge, 2002; Sanderson et al., 1994; Subramanian and Meyer, 1997), it is of vital importance to elucidate the molecular mechanisms involved in the ER/SR-stress associated drop in ER/SR Ca2+ content, so that therapeutic targets can be identified for this fundamental disease process. In summary, the present study shows that the causal linkage between the ER/SR stress response and Ca2+ dysregulation occurs before initiation of the UPR when ER/SR accumulation of unfolded protein is in its early stages.
