2013, Biotechnology AdvancesCitation Excerpt :Since gene–gene interaction typically happens inside a cell, analysis of the single-cell transcriptome is the most efficient method to reveal the regulatory relationship among genes. The small amount of RNA within a mammalian cell (~ 20–40 pg of total RNA (Uemura, 1980)) is a challenge for transcriptome analysis. Our group has overcome technical hurdles for single-cell molecular analysis by using microfluidic technology (Chen and Zhong, 2008; Fan et al., 2012; Zhong et al., 2008a).

2006, Progress in Nucleic Acid Research and Molecular BiologyCitation Excerpt :Unfortunately, we had not only to realize that the study is flawed due to inadequate receptor‐specific primers (275) but also figured out that the published variants of the TPEA design (273, 276) do not lead to proper preamplification. A mammalian cell contains on average between 20 and 40 pg total RNA (277, 278), which is equivalent to about 0.5–1.0 pg mRNA or 105–106 mRNA molecules. Handling such small RNA amounts is already challenging, but an uneven distribution of different RNA species (279) complicates the situation further.
