The influence of host and tissue age on life span and growth rate of serially transplanted mouse mammary gland

Abstract

Mouse mammary gland has a finite life span when serially transplanted in isogenic female hosts, using declining rate of ductal growth as an indicator of aging. In the experiments reported here, young and old hosts were transplanted with mammary tissues from young and old donors, and the transplants were propagated serially until they were lost due to diminished capacity for growth. Mammary tissue derived from both young and old donors behaved similarly when propagated in young hosts, and no consistent effect of donor age was observed. Old hosts, in contrast, did not support vigorous growth of either old or young grafts, probably because of endocrine insufficiency. The lack of vigorous growth during the first transplant generation in old hosts did not reduce the growth potential of either young or old transplants, since both grew well when retransplanted into young hosts. The results suggest that the life span of mammary cells is primarily related to a limited potential for division.

Résumé

La longévité des glandes mammaires de souris est limitée quand ces galndes sont transplantées sur des animaux femelles isogène et que l'on prend le ralentissement de la rapidité de croissance des conduits comme indicateur du viellisseement. Dans les expériences décrites ici, des tissues mammaires de donneurs jeunes et âgés ont été transplantés sur des récepteurs jeunes et âgés et les transplants ont été propagés en série jusqu'au moment où on les a perdus du fait de la diminution de leur faculté de développement. Les tissus mammaires de donneurs jeunes et vieux se sont comportés de façon similaire chez les récepteurs jeunes et aucun effet notable imputable à l'âge du donneur n'a été observé. Les récepteurs âgés n'ont, par contre et probablement à cause d'insufffisances endocriniennes, pas favorisé le développement vigoureux des greffons, jeunes ou âgés. L'absence de croissance vigoureuse de la première génération de transplants chez les récepteurs âgés n'a pas fait diminuer le potentiel de croissance des transplants, jeunes ou vieux, puisque les uns et les autres se développaien bien quand on les retransplantait chez des réceptteurs jeunes. Les résultats suggèrent que la longévité des cellules mammaires est en rélation primaire avec un potentiel de division limité.

Zusammenfassung

Die Brustdrüse der Maus hat eine begrenzte Lebensdauer bei Serientransplantation in isogene weibliche Empfänger. Als Altersindikator wird die Abnahme des Ductuswachstums herangezogen. In den hier beschriebenen Experimenten erhielten junge und alte Empfänger Brustdrs̈engewebe von jungen und alten Spendern. Die Transplantate wurden serienmäβig fortgeführt, bis der Verlust durch verminderte Wachstumskapazität eintrat. Brustdrüsengewebe von jungen und alten Tieren verhielt sich gleichsinnig in jungen Empfängern. Ein Effekt des Spenderalters konnte nicht festgestellt werden. Dazu im Gegensatz unterhielten alte Empfänger kein starkes Wachstum von jungen oder alten Transplantaten, wahrscheinlich wegen endokriner Insuffizienz. Das Fehlen starken Wachstums während der ersten Transplantationsgeneration bei alten Empfängern verminderte das Wachstumspotential der jungen und alten Transplantate nicht, da beide nach Retransplantation in junge Empfänger gut wuchsen. Die Ergebnisse deuten an, daβ Lebensdauer der Brustdrüsenzellen primär durch eine begrenzte Teilungspotenz beding ist.
