In contrast, M2 mAChRs are primarily found on cholinergic neurons and act as presynaptic autoreceptors to control ACh release (Zhang, Basile, et al., 2002, Zhang, Yamada, Gomeza, Basile, & Wess, 2002; Rouse, Edmunds, Yi, Gilmor, & Levey, 2000; Rouse, Gilmor, & Levey, 1998). This mAChR subtype is expressed throughout the CNS, specifically within the basal forebrain, parts of the striatum, cranial motor nuclei, non-pyramidal neurons of the hippocampus, and the pedunculopontine and laterodorsal tegmental nuclei (PPTg and LDTg, respectively) of the brainstem.

In this regard, M2 plays a major role in modulating cholinergic tone and broadly impacting circuit function (Levey et al., 1995a; Levey et al., 1995b; Rouse and Levey, 1996; Rouse et al., 1997, 2000). M2 receptors are also the most likely candidates to mediate the muscarinic suppression of GABA release in the hippocampus (Hájos et al., 1997; Lawrence, 2008; Shirey et al., 2008).
