ReviewInhibition of TNF-induced apoptosis by NF-κB
Abstract
Tumour necrosis factor (TNF) is an inflammatory cytokine possessing a unique property: it can induce cells to undergo apoptosis. The sensitivity of different cell types to TNF-induced apoptosis can vary dramatically, but most cells become very sensitive upon simultaneous treatment with inhibitors of protein synthesis. It has been suggested therefore that a gene, or set of genes, is induced upon TNF receptor activation that downregulates the apoptosis signal. Recent results have shown that NF-kB, a transcription factor activated upon TNF signalling, is at least partly responsible for this effect. These findings have broadened the role of NF-kB from that of a regulator of immune and inflammatory responses to include an involvement in the regulation of apoptosis.

Aiming at early-stage vulnerable plaques: A nanoplatform with dual-mode imaging and lipid-inflammation integrated regulation for atherosclerotic theranostics
2024, Bioactive MaterialsThe vulnerable plaques in atherosclerosis can cause severe outcome with great danger of acute cardiovascular events. Thus, timely diagnosis and treatment of vulnerable plaques in early stage can effectively benefit the clinical management of atherosclerosis. In this work, a targeting theranostic strategy on early-stage vulnerable plaques in atherosclerosis is realized by a LAID nanoplatform with X-CT and fluorescent dual-mode imaging and lipid-inflammation integrated regulation abilities. The iodinated contrast agents (ICA), phenylboronic acid modified astaxanthin and oxidized-dextran (oxDEX) jointly construct the nanoparticles loaded with the lipid-specific probe LFP. LAID indicates an active targeting to plaques along with the dual-responsive disassembly in oxidative stress and acidic microenvironment of atherosclerosis. The X-CT signals of ICA execute the location of early-stage plaques, while the LFP combines with lipid cores and realizes the recognition of vulnerable plaques. Meanwhile, the treatment based on astaxanthin is performed for restraining the progression of plaques. Transcriptome sequencing suggests that LAID can inhibit the lipid uptake and block NF-κB pathway, which synergistically demonstrates a lipid-inflammation integrated regulation to suppression the plaques growing. The in vivo investigations suggest that LAID delivers a favorable theranostics to the early-stage vulnerable plaques, which provides an impressive prospect for reducing the adverse prognosis of atherosclerosis.
Surveying the landscape of emerging and understudied cell death mechanisms

A large and growing number of mammalian cell death mechanisms have been described over the past few decades. Major pathways with established roles in normal or disease biology include apoptosis, necroptosis, pyroptosis and ferroptosis. However, additional non-apoptotic cell death mechanisms with unique morphological, genetic, and biochemical features have also been described. These mechanisms may play highly specialized physiological roles or only become activated in response to specific lethal stimuli or conditions. Understanding the nature of these emerging and understudied mechanisms may provide new insight into cell death biology and suggest new treatments for diseases such as cancer and neurodegeneration.
Protective effects of metformin against aluminum phosphide-induced acute hepato-renal damage in rats: An experimental approach
2022, Pesticide Biochemistry and PhysiologyPhosphine (PH3), from hydrolysis of magnesium, zinc, and aluminum phosphide (AlP), is a rodenticide and insecticide which is used to avoid losses of the agriculture products. However, using of this agent may affect the human health, in a way that poisoning with AlP has a high rate of mortality and morbidities. This study determined the ameliorative effects of metformin (MET) on AlP-induced hepato- and nephro-toxicity in Wistar rats. Male rats were randomly divided into four experimental groups. Group I was the control group received coconut oil by oral gavage, group II was the model group received AlP (12 mg/kg) distributed in coconut oil by oral gavage, group III received MET (200 mg/kg; i.p.), and group IV received MET (200 mg/kg; i.p.) 30 min after intoxication. After 24 h, the serum, liver and kidney tissues were collected for histopathological and biochemical investigations. The levels of kidney function markers, blood urea nitrogen and creatinine, and liver function markers, ALP, AST and ALT, in the plasma were increased significantly followed by AlP intoxication. The results revealed that phosphine causes a significant enhancement of lipid peroxidation, while decreases the activity of superoxide dismutase in both liver and kidney tissues. Furthermore, phosphine significantly induced the up-regulation of TNF-α and phosphorylation of NF-κB in target tissues. Overall, treatment with MET abolished aforementioned alterations resulted by AlP intoxication. Furthermore, histological evaluation indicated a deleterious effect of AlP on the liver and kidney tissues along with marked increase in kidney and liver injury scores, which is mitigated by MET administration. According to our results, although metformin could not bring the changes to the level of the control group, it was indicated that this drug might possess a protective effect against AlP-induced hepato and nephrotoxicity by inhibiting inflammatory responses and oxidative stress.
Toxicological impacts of nanopolystyrene on zebrafish oocyte with insight into the mechanism of action: An expression-based analysis
2022, Science of the Total EnvironmentMany studies have investigated the negative impacts of microplastics on teleost fishes with very little or no evidence of their mechanism of action. This scenario entreats us to investigate the toxicities of nanopolystyrene in zebrafish oocyte with emphasis on the mechanism of action. In the present study, the cellular levels of mRNA transcripts of different genetic markers (such as: sod, gpx, nrf2, inos, ucp2, and atp6 (redox-sensitive markers); nfkβ, tnfα, il-10, ikβ, gdf9, and bmp15 (immune markers); gadd45, rad51, p53 and bcl2 (DNA damage and apoptotic)) have been quantified by real-time PCR after 6 h of incubation of isolated oocyte with different doses of nanopolystyrene viz. P0 (control i.e. no polystyrene in culture medium), P1 (100 ng/ml), and P2 (400 ng/ml). Results showed that both the treatment concentrations of nanopolystyrene induce oxidative stress with % DPPH = 30.75, 31.61, and 32.43% for P0, P1, and P2, respectively. Increase in oxidative stress in oocytes with increasing doses of nanopolystyrene was also observed in TBARS assay with MDA content 0.12 and 0.21 μM for P1 and P2, respectively as compaired to the control 0.08 μM. This increased oxidative stress can regulate the expression pattern (upregulation/downregulation) of selected genes leading to different toxic effects like – oxidative stress, immunotoxicity, and apoptosis in oocytes, which suggests the impairment of reproductive functions by nanopolystyrene.
Impacts of DigestaWell NRG Supplementation on Post Exercise Muscle Soreness in Unconditioned Horses, a Pilot Study
2021, Journal of Equine Veterinary ScienceExercising horses are commonly plagued by muscle fatigue and soreness, which can result in reduced performance ability. In the present study, ten unconditioned horses were fed 200g per day DigestaWell NRG, a commercial dietary supplement containing Yucca schidigera and Trigonella foenum-graecum, two herbs shown in other species to reduce post-exercise muscle pain and soreness. A control, unsupplemented group contained ten horses of similar age, breed, and gender. Horses completed a 50 minutes, ridden standardized exercise test of moderate intensity immediately prior to (Period1) and after 28 days of supplementation (Period2). Muscle soreness and tightness were evaluated 24 hours prior to and after each exercise test and used to determine the percent increase in post-exercise muscle soreness and tightness. Blood samples were collected before, and at 10 and 30 minutes, and 1, 4, and 24 hours post exercise. Plasma was analyzed for glucose, lactate, non-esterified fatty acid, tumor necrosis factor-α, and interleukin-1β concentrations. Data were analyzed by repeated measures ANOVA using SAS Enterprise Guide v. 7.1. No changes in plasma parameters were indicated between periods for unsupplemented horses (P > 0.1) during Period2, excepting glucose, which was greater during Period2 (P = 0.018). Supplemented horses had lesser concentrations of tumor necrosis factor-α (P = 0.016) and lactate (P = 0.058) during Period2 than during Period1. During Period2, supplemented horses experienced a smaller percent increase in post exercise muscle soreness (P = 0.031). DigestaWell NRG supplementation may benefit unconditioned horses undergoing moderate intensity exercise through reducing lactate production and inflammation.
Characterization of sediment toxicity in Shanghai Harbor using toxicity tests and digital gene expression analysis based on clams Ruditapes phillipinarum
2020, Ecotoxicology and Environmental SafetyIn the present study, chemical analysis of contaminants (three classes of organic pollutants and seven metals) and elutriate toxicity test were adopted to evaluate the potential environmental hazards of dredged sediment samples from five sites (SS1-5) along Huangpu River Channel (Shanghai Harbor, China). The metal Pb, Cu, Cr, Zn and the organic pollutants including total hexachlorocyclohexane (HCHs) and total dichlorodiphenyltrichloroethane (DDTs) in the five samples exceeded the threshold for effects level (TEL) to varying degrees. The probable effect concentration quotients (QPECm) of contaminants from the five dredged samples were all above 0.25, which means potential toxicity risks. Elutriate toxicity tests using medaka fish (Oryzias melastigma) and manila clam (Ruditapes philippinarum) showed that SS2 caused mortality to both species and SS1 caused mortality to fish. To explore the molecular biomarkers that may reflect the toxic effects, differential expressed genes were identified by RNA-Seq-based transcriptome profiling from the survived clams exposed to the two polluted elutriates (SS1, SS2). In clams exposed to SS1 and SS2 elutriate, 368 and 860 differential expressed genes (DEGs) were up-regulated, 199 and 1304 genes were down-regulated, respectively. Fourteen DEGs were selected from the enriched pathways that reflect cytotoxicity and responses to xenobiotics for the following quantitative real time PCR analysis. The transcriptomic profiling and the selected gene's expression patterns from clams exposed to SS1 and SS2 showed significant differences with the non-contaminated and control groups. Using the expression data of the selected gene battery in Factor Analysis allowed the discrimination between contaminated and non-contaminated sites and may reflect an influence gradient of sites. The development of the assay of these molecular biomarkers may provide a rapid and high-throughput tool for the quality assessment of the dredging sediments.
