Effect of the phenylpyrrole fungicide fludioxonil on cell proliferation and cardiac differentiation in mouse embryonic stem cells
2021, Reproductive ToxicologyCitation Excerpt :These mEBs can initiate differentiation into the three germ layers, the ectoderm, mesoderm, and endoderm. Hence, mEBs play an important role in the differentiation system of mESCs, and the quality of mEBs affects the subsequent differentiation [6]. The heart is the first functional embryonic organ that develops in the mesoderm after gastrulation, the process by which the mesoderm is formed from the ectoderm [7].
Pre-validation study of alternative developmental toxicity test using mouse embryonic stem cell-derived embryoid bodies
2019, Food and Chemical ToxicologyCitation Excerpt :These methods consume a lot of time and effort moreover there can be large errors depending on the experimenter performing the test. The embryoid bodies test (EBT), a developmental toxicity testing method, is a cytotoxicity test that evaluates reductions in embryoid body (EB) area in order to determine drug toxicity (Ohnuki and Kurosawa, 2013; Zhou et al., 2010). The EBT measures the reduction of EB area as an endpoint after 3 days of chemical exposure and avoids problems associated with conventional cytotoxicity tests.
Advanced developmental toxicity test method based on embryoid body's area
2017, Reproductive ToxicologyCitation Excerpt :EBs with embryo-like structures act as the onset point for in vitro differentiation, recapitulate many features of differentiation during early mammalian embryogenesis, and therefore are useful as in vitro model systems for the study of mammalian embryogenesis. Since the quality of EBs affects the induction efficiency of EB derivatives in subsequent differentiation culture [4], a decrease in size or quality of EBs due to exposure to chemicals can result in growth retardation and abnormal differentiation during embryo development. The mechanism related to decrease in EB area due to toxicants was investigated by analyzing differentially expressed genes (DEG) via NGS.
Biomaterials control of pluripotent stem cell fate for regenerative therapy
2016, Progress in Materials ScienceCitation Excerpt :The combination of collagen and Matrigel was shown to provide the necessary components of ECMs as well as physical supports for ESC development and differentiation. The mouse ESCs seeded within the 3D gel of collagen/Matrigel grew into aggregates and formed embryoid bodies, which eventually differentiated into three embryonic germ layers after 7 days of culture [256]. Moreover, the human ESCs differentiated into cardiomyocytes with the introduction of ascorbic acid and demonstrated the beating behavior at 19 days.
