DNA synthesis and differentiation in developing white adipose tissue

Abstract

The relationship between proliferation and differentiation was investigated during the development of subcutaneous white adipose tissue of rats aged from the 22nd day of gestation to the 3rd day after birth. Pulse-labeling with thymidine-3H was followed by combining histochemical visualization of α-naphthyl acetate esterase activity and autoradiography on the same section. Cells of so-called fat organs were divided into three categories on the basis of the esterase reaction: undifferentiated mesenchymal cells, partly differentiated preadipocytes, and fully differentiated adipocytes. Proliferative activity was assessed by measuring labeling indices of the three categories of developing fat cells. In addition, overall labeling indices were determined as a function of the ratio of differentiated to undifferentiated cell forms. Labeling indices were highest in partly differentiated cells or fat organs. The results suggest a functional participation of cell replication in the control of the differentiation process.

      Unexpectedly, a terminal adipocyte differentiation marker, Perilipin1 (PLIN1), was also expressed in numerous EdU flash-labeled (i.e., 2 hr exposure) cells, especially after 3 days of CL treatment. EdU+ adipocytes were small (<10 μm) and contained small lipid droplets, indicating that newly differentiating adipocytes can divide (Pilgrim, 1971). ( We have not observed cell division in mature, unilocular adipocytes.)
